regulator of mRNA translation; Specifically recognizes and binds N6-methyladenosine (m6A)-containing mRNAs, and promotes mRNA translation efficiency. promotes ribosome loading to m6A-containing mRNAs and interacts with translation initiation factors eIF3 (EIF3A or EIF3B) to facilitate translation initiation. Required to facilitate learning and memory formation in the hippocampus by enhancing protein synthesis upon neuronal stimulation. Acts as a negative regulator of antigen cross-presentation in myeloid dendritic cells.

knockdown of YTHDF1 in Merkel cell carcinoma (MCC) cell lines negatively affected the translation initiation factor eIF3 and reduced proliferation and clonogenic capacity in vitro. YTHDF1 deficiency inhibits NSCLC cell proliferation and xenograft tumor formation through regulating the translational efficiency of CDK2, CDK4, and cyclin D1, and YTHDF1 depletion restrains de novo lung adenocarcinomas (ADC) progression. Knocking down the expression of YTHDF1 significantly inhibited the colorectal cancer (CRC) cell's tumorigenicity in vitro and murine xenograft tumor growth in vivo.
