YTHDC2 und shRNA*
CAHL; hYTHDC2
Homo sapiens
RNA Helicase; 3'-5' RNA helicase that plays a key role in the male and female germline by promoting transition from mitotic to meiotic divisions in stem cells. Specifically recognizes and binds N6-methyladenosine (m6A)-containing RNAs, a modification present at internal sites of mRNAs and some non-coding RNAs that plays a role in the efficiency of RNA processing and stability.
ohne Gefährdungspotential
CRISPR-Cas9 mediated YTHDC2 knockout notably inhibited GC (gastric cancer) cell viability, proliferation and invasion. Using a xenograft tumor model, knockout of YTHDC2 markedly reduced tumor size and lung metastasis nodules in vivo. The in vivo and in vitro studies revealed that YTHDC2 overexpression inhibited the proliferation and migration of lung cancer cells as well as tumor growth in nude mice. Knocking down YTHDC2 promoted cell cycle progression by accelerating the progression of cells from the G1 to the S phase in BEAS-2B and H1299 cells. EdU assay showed that the proliferation of BEAS-2B and H1299 cells was significantly enhanced by knocking down YTHDC2. Die ursächliche Beteiligung an der Entstehung von Tumoren ist nicht gezeigt.
