E-cadherin is frequently mutated in several types of cancers. The functional loss of cell adhesion and cell junctions mediated by loss of E-cadherin homophilic binding enables cells to dissociate from the primary tumor, invade surrounding tissues and migrate to distant sites and establish metastatic tumors. Soluble E‐cad (sE‐cad), which is the extracellular proteolytic fragment of E‐cad, promotes tumor cell invasion and metastasis by stimulating the activity of multiple matrix metalloproteinases (MMPs) and a disintegrin and metalloproteases (ADAMs). sE‐cad binds to surface receptors in an autocrine or paracrine manner to promote cancer cell migration, proliferation and survival by activating or inhibiting multiple signaling pathways.
